Biological Background: β-Catenin Function and Localization
- β-Catenin (CTNNB1) acts as a key downstream transcriptional coactivator in the canonical Wnt signaling pathway, driving expression of TCF/LEF target genes. Related references: PMID:17524503 PMID:18077326 PMID:18086858
- In the absence of Wnt ligand, β-Catenin is targeted for degradation by a destruction complex containing AXIN, APC, CSNK1A1, and GSK3B, which phosphorylates it for ubiquitination by BTRC. Related references: PMID:17524503 PMID:18077326 PMID:18086858
- It also serves as a structural component of cadherin-based adherens junctions, linking E-cadherin to the actin cytoskeleton and regulating cell adhesion.
- β-Catenin shuttles between the cytoplasm and nucleus, and is also localized at cell membranes, adherens junctions, centrosomes, and spindle poles.
- Post-translational modifications include phosphorylation, acetylation, S-nitrosylation, and sumoylation, which regulate its stability and signaling activity.
- Expressed in hair follicles, colon epithelia, and cortical neurons, with elevated levels often observed in breast cancer tissues. Related references: PMID:29367600
- Disease-associated mutations in CTNNB1 are linked to intellectual disability, chromosomal rearrangements, and various cancers.
Experimental Guidance and Technical Tips
- For Western blot, expect a band at ~85 kDa. Validate in Wnt-activated versus resting cell lysates to confirm specificity.
- For immunohistochemistry (IHC-P), antigen retrieval may be required depending on fixation conditions. Optimize retrieval buffer and time for the target tissue.
- For immunofluorescence/ICC, consider using cells treated with Wnt agonists or LiCl to enhance nuclear β-Catenin signal.
- The antibody may recognize total β-Catenin irrespective of its association with cadherins or other proteins; co-immunoprecipitation assays require separate validation.
- Since the immunogen corresponds to the N-terminal region (aa 1–100), the antibody may detect both full-length and certain truncated forms; confirm band patterns with controls.
