Biological Background: β-arrestin1 Function and Localization
- β-arrestin-1 regulates agonist-mediated GPCR signaling by mediating receptor desensitization and resensitization processes. Related references: PMID:37209686 PMID:38175886 PMID:40384633
- It also functions as an endocytic adapter, targeting many receptors for internalization via clathrin-coated pits, and can act as a signaling scaffold for MAPK, AKT1, and p38 pathways.
- Upon GPCR activation, β-arrestin-1 can translocate to the nucleus and regulate transcription by recruiting histone acetyltransferases to promoter regions.
- It negatively regulates the NOTCH signaling pathway by mediating ITCH-dependent ubiquitination and degradation of NOTCH1. Related references: PMID:23886940
- It participates in MRGPRE-mediated signaling to improve glucose tolerance by promoting ALDOA phosphorylation, increasing glycolysis and GLP-1 secretion. Related references: PMID:40446798
- The protein is localized in the cytoplasm, nucleus, cell membrane, clathrin-coated pits, pseudopodia, and cytoplasmic vesicles, reflecting its dynamic role in receptor trafficking.
- β-Arrestin-1 is subject to phosphorylation and ubiquitination, and it exists in alternatively spliced isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-100) of human β-arrestin1, which is conserved in mouse; consider using this antibody for detecting endogenous β-arrestin1 in human and mouse cell lysates by Western blot.
- For Western blot, whole-cell lysates or tissue homogenates can be prepared using standard RIPA buffer; validate the antibody in the relevant sample system as expression levels and post-translational modifications may affect detection.
- The antibody is suitable for ELISA; optimize coating antigen concentration and detection parameters for your specific assay format.
