Biological Background: β-Amyloid(1-40) / APP Function and Localization
- APP is a type I transmembrane protein that functions as a cell surface receptor, promoting neurite outgrowth, neuronal adhesion, and synaptogenesis. Interaction between APP molecules on adjacent cells facilitates synapse formation. Related references: PMID:25122912
- Acts as a kinesin I membrane receptor, mediating the axonal transport of beta-secretase and presenilin 1, and plays a role in anterograde transport of cargo towards synapses. Related references: PMID:17062754, PMID:23011729
- Proteolytic processing of APP generates amyloid-beta peptides, including β-Amyloid(1-40) and (1-42), which are lipophilic metal chelators with metal-reducing activity, binding transient metals like copper, zinc, and iron. These peptides can inhibit metal-catalyzed oxidation of lipoproteins.
- β-Amyloid(1-42) is a more effective reductant than β-Amyloid(1-40) and may activate mononuclear phagocytes, eliciting inflammatory responses in the brain.
- The gamma-CTF peptides and caspase-cleaved C-terminal fragments, such as C31, are potent enhancers of neuronal apoptosis.

Experimental Guidance and Technical Tips
- The immunogen used corresponds to amino acids 671-770 of human APP (NP_000475.1), a region encompassing the C-terminal portion of the amyloid-beta peptide and beyond. This antibody may recognize full-length APP, C-terminal fragments, and the free β-Amyloid(1-40) peptide; validate specificity in your experimental system.
- For immunofluorescence (IF-P) and immunohistochemistry (IHC-P), consider optimizing fixation and permeabilization conditions. Antigen retrieval may be necessary for paraffin-embedded tissues.
- In dot blot (DB) and ELISA applications, ensure the peptide is properly coated or immobilized. The antibody’s performance may depend on the conformation and purity of the peptide sample.
- Cross-reactivity with mouse enables testing in mouse models of Alzheimer's disease; however, confirm reactivity in your specific strain and tissue.
