Biological Background: UVRAG Function and Localization
- UVRAG (UV radiation resistance-associated gene protein), also known as p63, is a versatile regulator of membrane trafficking, including COPI-dependent retrograde transport from Golgi and autophagy, by associating with the NRZ complex and PI3K complex II. Related references: PMID:16799551, PMID:18552835, PMID:20643123, PMID:24056303
- It activates the lipid kinase activity of PIK3C3 and is essential for autophagosome maturation, endocytosis, and ATG9A trafficking. Related references: PMID:16799551, PMID:24056303
- UVRAG promotes late endocytic membrane fusion by enhancing class C Vps/HOPS complex association with SNAREs and facilitating Rab7 activation. Related references: PMID:18552835, PMID:28306502, PMID:24550300
- In the nucleus, UVRAG maintains chromosomal stability by promoting DNA double-strand break repair through non-homologous end joining (NHEJ) via interaction with the DNA-PK complex. Related references: PMID:22542840
- It localizes to multiple subcellular compartments including late endosomes, lysosomes, autophagosomes, early endosomes, endoplasmic reticulum, midbody, and centromeres.
- Highly expressed in brain, lung, kidney, and liver, highlighting its importance in these tissues.
- UVRAG contains a coiled coil domain and undergoes phosphorylation; it is involved in viral entry for negative-strand RNA viruses by promoting endocytic transport and VAMP8-specific SNARE assembly. Related references: PMID:24550300
Experimental Guidance and Technical Tips
- For Western blot (WB), consider using 30–50 µg of total protein lysate from human, mouse, or rat cell lines or tissues, given the predicted molecular weight of ~78 kDa.
- For immunohistochemistry (IHC-P), antigen retrieval may be required; validate in appropriate positive control tissues such as brain or liver where UVRAG is highly expressed.
- The immunogen corresponds to amino acids 501–699 of human UVRAG; ensure that the detected band aligns with the expected size and appears in control samples.
- Cross-reactivity is reported for human, mouse, and rat; for other species, sequence homology analysis is recommended.
- This polyclonal antibody may be suitable for ELISA under optimized conditions; perform titration to determine the optimal working concentration.
