Biological Background: UTP11L Function and Localization

  • Probable U3 small nucleolar RNA-associated protein 11, also known as UTP11-like protein (UTP11L), is encoded by the UTP11 gene in humans. Alternative names include CGI94 and CGI-94.
  • It is a component of the small subunit (SSU) processome, the earliest precursor of the small ribosomal subunit (40S). During ribosome biogenesis, the SSU processome assembles in the nucleolus and facilitates pre-rRNA folding, modification, cleavage, and assembly.
  • UTP11L is specifically involved in nucleolar processing of pre-18S ribosomal RNA, a critical step in the maturation of the small ribosomal subunit.
  • Subcellularly, UTP11L localizes to the nucleolus, the site of active rRNA transcription and early ribosome assembly steps.
  • The protein is subject to post-translational modifications including phosphorylation and isopeptide bond formation (ubiquitination), and undergoes alternative splicing.
  • As a reference proteome member with a molecular weight of approximately 30.4 kDa, UTP11L serves as a marker for nucleolar function and ribosome biogenesis research.

Experimental Guidance and Technical Tips

  • The immunogen is designed against a central region (aa 110–240) of human UTP11L, which is conserved in mouse and rat, supporting cross-reactivity. Confirm sequence identity in your target species if needed.
  • For Western blot, a band around 30 kDa is expected. Consider using appropriate positive and negative controls to validate specificity.
  • For ELISA, optimize coating concentration and detection antibody dilution. Validate in the relevant sample matrix (e.g., human, mouse, or rat lysates).
  • Given the nucleolar localization, protein extraction may benefit from nuclear enrichment protocols to improve detection sensitivity.
  • As a polyclonal antibody, lot-to-lot consistency should be verified for critical applications.
