Biological Background: USP9X Function and Localization
- USP9X encodes a deubiquitinase that processes ubiquitin precursors and removes ubiquitin from proteins, modulating protein turnover and preventing degradation.
Related references: PMID:18254724 PMID:19135894 PMID:22371489 - The enzyme selectively hydrolyzes Lys-11-, Lys-63-, Lys-48-, and Lys-6-linked polyubiquitin chains.
Related references: PMID:30914461 - USP9X is an essential component of TGF-β/BMP signaling, deubiquitinating monoubiquitinated SMAD4, opposing the E3 ligase TRIM33.
Related references: PMID:19135894 - It stabilizes RICTOR by removing Lys-63-linked polyubiquitin chains, promoting mTORC2 complex assembly and kinase activity.
Related references: PMID:33378666 - USP9X regulates chromosome alignment and segregation during mitosis by controlling BIRC5/survivin localization to centromeres.
Related references: PMID:16322459 - It enhances circadian clock function by deubiquitinating and stabilizing BMAL1.
Related references: PMID:29626158 - Subcellularly, USP9X resides in the cytosol, growth cones, and cilium axoneme, consistent with roles in neuronal growth and ciliary function.
- The protein is widely expressed in embryonic and adult tissues and is implicated in intellectual disability (XLID99) when mutated.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide corresponding to amino acids 1–100 of human USP9X. Consider using Western blot (WB) or ELISA as primary detection methods; always validate antibody performance in your specific sample system.
- USP9X is a high molecular weight protein (~290 kDa). For WB, use a low-percentage polyacrylamide gel (e.g., 6–8%) and prolonged transfer times to ensure efficient detection.
- Cross-reactivity is reported with Human, Mouse, and Rat; however, verify species compatibility in your experimental context.
- The antibody is supplied as a rabbit polyclonal; therefore, batch-to-batch consistency should be evaluated for quantitative assays.
