Biological Background: USP39 Function and Localization

  • Ubiquitin carboxyl-terminal hydrolase 39 (USP39), also known as SAD1 homolog and U4/U6.U5 tri-snRNP-associated 65 kDa protein, is a deubiquitinating enzyme containing a zinc-finger motif.
  • Essential component of the U4/U6-U5 tri-snRNP complex, playing a critical role in pre-mRNA splicing and spliceosome assembly. Related references: PMID:11350945 PMID:26912367
  • Regulates AURKB mRNA levels, thereby contributing to cytokinesis and the spindle checkpoint during cell division. Related references: PMID:18728397
  • Functions in DNA damage response by deubiquitinating and stabilizing CHK2, regulating apoptosis and G2/M cell cycle arrest. Related references: PMID:30771428
  • Facilitates DNA double-strand break repair by controlling XRCC4/LIG4 recruitment for non-homologous end-joining. Related references: PMID:34614178
  • Exhibits antiviral activity through stabilization of STAT1 and modulation of type I IFN signaling. Related references: PMID:33127822
  • Negative regulator of NF-kappa-B activation by deubiquitinating and stabilizing NFKBIA. Related references: PMID:36651806
  • Localized in the nucleus, USP39 is a phosphoprotein involved in alternative splicing, ubiquitin conjugation, and metal-binding pathways.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide within amino acids 200-300 of human USP39. Consider this epitope region when designing complementary reagents or performing validation studies.
  • For Western blotting (WB), anticipate a band at approximately 65 kDa. Validate detection in human and rat lysates, as the antibody shows cross-reactivity with both species.
  • In immunohistochemistry (IHC-P), nuclear staining is expected due to USP39’s subcellular localization. Antigen retrieval methods may be required; optimize according to tissue type.
  • ELISA protocols can be developed with standard direct or sandwich formats; verify linearity and sensitivity using recombinant USP39 or cell lysates.
