Biological Background: USP22 Function and Localization

  • USP22 is a ubiquitin carboxyl-terminal hydrolase (deubiquitinase) also known as USP3L, encoded by the USP22 gene (KIAA1063, USP3L).
  • As part of the SAGA histone acetyltransferase complex, USP22 catalyzes the deubiquitination of histones H2A and H2B, functioning as a transcriptional coactivator and being recruited by MYC to gene promoters. Related references: PMID:18206972, PMID:18206973, PMID:18469533
  • USP22 promotes cell cycle progression by stabilizing cyclin B1 (CCNB1) and protecting it from proteasome-mediated degradation. Related references: PMID:27030811
  • Through stabilization of the deacetylase SIRT1, USP22 antagonizes TP53 transcriptional activation, thereby modulating cell cycle and apoptosis. Related references: PMID:22542455

  • The immunogen (N-terminal amino acids 1-175) is well suited for detecting full-length USP22; consider using nuclear and cytoplasmic extracts to verify subcellular localization in WB.
  • For Western blot, the predicted molecular weight is approximately 60 kDa; include appropriate positive controls and validate in your sample of interest.
