Biological Background: USP21 Function and Localization

  • USP21 (Ubiquitin carboxyl-terminal hydrolase 21) is a deubiquitinating enzyme belonging to the USP family, also known as USP16, USP23, and deubiquitinating enzyme 21.
  • It deubiquitinates histone H2A, a specific tag for epigenetic transcriptional repression, thereby acting as a transcriptional coactivator. Deubiquitination of H2A relieves repression of H3K4 di- and trimethylation, regulating transcriptional initiation (By similarity).
  • USP21 deubiquitinates BAZ2A/TIP5, leading to its stabilization and involvement in epigenetic regulation. Related references: PMID:26100909
  • It can remove NEDD8 from NEDD8 conjugates but has no effect on Sentrin-1 conjugates. Related references: PMID:10799498
