Biological Background: USP15 Function and Localization
- USP15 (ubiquitin carboxyl-terminal hydrolase 15), also known as UNPH4 or UNPH-2, is a deubiquitinating enzyme that cleaves ubiquitin chains from target proteins, and is expressed as multiple isoforms with observed molecular weights of ~27 kDa and 109–112 kDa due to alternative splicing.
- Functions as a critical regulator of TGF-beta receptor signaling by deubiquitinating monoubiquitinated R-SMADs (SMAD1/2/3) or stabilizing TGFBR1, thereby promoting TGF-beta target gene activation. Related references: PMID:21947082, PMID:22344298
- Modulates the NF-kappa-B pathway by mediating deubiquitination of NFKBIA within the COP9 signalosome complex, linking it to transcriptional regulation. Related references: PMID:16005295, PMID:17318178, PMID:19576224
- Acts as an inhibitor of mitophagy by counteracting parkin (PRKN)-mediated ubiquitination; hydrolyzes Lys-48- and Lys-63-linked polyubiquitin chains on mitochondrial substrates such as MFN2. Related references: PMID:24852371
- Deubiquitinates histone H2B, thereby influencing gene expression and DNA repair processes. Related references: PMID:24526689
- Participates in endosome organization by deubiquitinating SQSTM1/p62, promoting vesicle transport to the cell periphery. Related references: PMID:27368102
- Negatively regulates antifungal immunity through Lys-27-linked deubiquitination of CARD9, leading to its inactivation. Related references: PMID:33093067
- Localizes to the cytoplasm, nucleus, and mitochondrion, and is predominantly expressed in skeletal muscle, kidney, heart, placenta, liver, thymus, lung, and ovary.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal peptide (amino acids 968–981) of human USP15, designed for specific recognition of both full-length and shorter isoforms.
- For Western blotting, expect predominant bands at ~27 kDa and 109–112 kDa; validate detection in the relevant sample system and optimize blocking/dilution conditions.
- This antibody is suitable for immunoprecipitation; consider using cell or tissue lysates expressing USP15 and confirm co-precipitation with known interaction partners.
- In ELISA applications, carefully titrate the coating antigen and antibody concentration to achieve optimal signal-to-noise ratios.
