Biological Background: USP13 Function and Localization

  • USP13, also known as Isopeptidase T-3 (ISOT-3) or Ubiquitin carboxyl-terminal hydrolase 13, is a deubiquitinase that removes ubiquitin from target proteins, regulating processes such as autophagy, ERAD, cell cycle progression, and DNA damage response. Related references: PMID:21571647 PMID:32772043 PMID:33592542
  • It is a key component of a regulatory loop controlling autophagy by deubiquitinating and stabilizing BECN1 and VPS34, thereby promoting autophagosome formation. Related references: PMID:32101753
  • USP13 participates in endoplasmic reticulum-associated degradation (ERAD) by deubiquitinating SKP2 and UBL4A, ensuring proper protein quality control in the ER.
  • It regulates cell cycle progression by stabilizing cell cycle proteins such as SKP2 and AURKB. Related references: PMID:32772043
  • The protein is involved in the DNA damage response and maintenance of genomic stability via deubiquitination of RAP80 and TOPBP1. Related references: PMID:33592542
  • USP13 positively regulates type I and type II interferon signaling by deubiquitinating STAT1, but negatively regulates antiviral response via STING1 deubiquitination. Related references: PMID:23940278 PMID:28534493
  • Subcellularly, USP13 localizes to the cytoplasm.
  • It is highly expressed in ovary and testes, and contains zinc-finger domains and a thiol protease active site.

Experimental Guidance and Technical Tips

  • The immunogen used to generate this antibody corresponds to amino acids 90–200 of human USP13, a region within the N-terminal portion of the protein. This design may offer specificity for USP13 over other USP family members.
  • For western blot (WB), a band at approximately 97 kDa is expected. Validate the antibody in relevant cell or tissue lysates, and consider using protease inhibitors during sample preparation to preserve USP13 detection.
  • This antibody shows cross-reactivity with mouse and rat, enabling its use in rodent model studies. Perform a pilot experiment to confirm reactivity in your specific sample type.
  • For ELISA, optimize the coating antigen concentration and antibody dilution to achieve the desired signal-to-noise ratio.
