Biological Background: USP1 Function and Localization
- USP1 is a deubiquitinating enzyme (DUB) also known as Ubiquitin carboxyl-terminal hydrolase 1, Deubiquitinating enzyme 1 (hUBP), and Ubiquitin thioesterase 1.
- It negatively regulates the Fanconi anemia DNA repair pathway by deubiquitinating monoubiquitinated FANCD2. Related references: PMID:15694335
- USP1 also participates in PCNA-mediated translesion synthesis (TLS) by removing ubiquitin from monoubiquitinated PCNA. Related references: PMID:16531995, PMID:20147293
- The enzyme has low intrinsic catalytic activity and requires the interaction with its cofactor WDR48 for high deubiquitinase activity. Related references: PMID:18082604, PMID:26388029
- Subcellularly, USP1 localizes to the nucleus, consistent with its role in DNA repair processes.
- USP1 belongs to the peptidase C19 family of ubiquitin-specific proteases and is a thiol protease.
- It is implicated in the ubiquitin-proteasome system and is subject to autocatalytic cleavage and phosphorylation.
- Given its role in DNA damage response, USP1 expression is critical for genome maintenance.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (amino acids 1-280); consider using this antibody to detect full-length USP1 or N-terminal fragments.
- For Western blot, optimize nuclear lysis and protein extraction conditions, as USP1 is a nuclear protein.
- Validate antibody specificity using positive and negative controls (e.g., USP1 knockout or siRNA knockdown samples).
- For ELISA assays, titrate the coating antigen and antibody dilution using recombinant USP1 or cellular lysates.
- Use appropriate anti-rabbit secondary antibodies for detection; ensure no cross-reactivity with target species reagents.
- In functional studies, co-expression of WDR48 may be required to observe USP1 enzymatic activity.
