Biological Background: UQCRFS1 Function and Localization
- UQCRFS1, also known as Rieske iron-sulfur protein (RISP), is a core subunit of the mitochondrial cytochrome b-c1 complex (Complex III) of the electron transport chain.
- It contains a [2Fe-2S] cluster and facilitates electron transfer from ubiquinol to cytochrome c1, contributing to the proton gradient via the Q cycle. Related references: PMID:31883641
- UQCRFS1 undergoes proteolytic processing upon insertion into the Complex III dimer, releasing a fragment called subunit 9, which is essential for proper complex assembly. Related references: PMID:28673544
- Incorporation of UQCRFS1 is the penultimate step in Complex III assembly; unprocessed fragments can disrupt complex function. Related references: PMID:28673544
- The protein is located in the mitochondrial inner membrane.
- Key UniProt keywords include 2Fe-2S, iron-sulfur, electron transport, respiratory chain, and primary mitochondrial disease.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 141–274 of human UQCRFS1, a region within the mature protein; the antibody is predicted to detect processed forms.
- In Western blot, a predominant band near 30 kDa is expected; additional bands representing proteolytic fragments may also be observed. Consider using mitochondrial-enriched lysates for enhanced signal.
- For IHC-P, antigen retrieval may be required; co-staining with a mitochondrial marker can help confirm localization.
- Cross-reactivity with mouse and rat is based on sequence homology; validation in the relevant species and sample type is recommended.
