Biological Background: UQCRC1 Function and Localization
- UQCRC1, also known as Cytochrome b-c1 complex subunit 1, mitochondrial, or Core protein I, is a component of the ubiquinol-cytochrome c oxidoreductase (complex III) in the mitochondrial respiratory chain.
- It participates in electron transfer from ubiquinol to cytochrome c, coupling this reaction to proton translocation across the mitochondrial inner membrane, which drives ATP synthesis.
- UQCRC1 and UQCRC2 are homologous to the mitochondrial-processing peptidase (MPP) subunits; UQCRC1 may retain MPP activity and is potentially involved in processing UQCRFS1 into the mature Rieske protein.
- Subcellular localization is the mitochondrial inner membrane.
- Tissue expression includes brain (substantia nigra, striatum, cortex, cerebellum), spinal cord, heart, kidney, liver, and muscle.
- Post-translational modifications include acetylation and phosphorylation.
- Genetic variants of UQCRC1 have been linked to neuropathy and Parkinsonism.
- UQCRC1 seems to play an important role in the maintenance of proper mitochondrial function in nigral dopaminergic neurons, with implications for Parkinson's disease (PubMed:33141179). Related references: PMID:33141179
- The computed molecular weight from sequence is ~52.6 kDa; the antibody detects a band at approximately 53 kDa in Western blotting.
Experimental Guidance and Technical Tips
- The immunogen was designed using a recombinant fusion protein spanning amino acids 156-480 of human UQCRC1; consider this antigen region when assessing epitope specificity.
- This antibody is suitable for applications including WB, IHC-P, IF/ICC, and ELISA; validate conditions in your specific sample matrix for optimal results.
- Reported cross-reactivity includes human, mouse, and rat; confirm in the relevant species model before use.
- As UQCRC1 is a mitochondrial inner membrane protein, consider using appropriate lysis and sample preparation methods to ensure proper solubilization and detection.
