Biological Background: UQCRC1 Function and Localization
- UQCRC1, also known as Cytochrome b-c1 complex subunit 1 or Core protein I, is a key component of the mitochondrial ubiquinol-cytochrome c oxidoreductase (Complex III), part of the electron transport chain that drives oxidative phosphorylation.
- Complex III catalyzes electron transfer from ubiquinol to cytochrome c and contributes to the proton gradient across the inner mitochondrial membrane through the Q cycle, coupling redox reactions to proton translocation.
- The core subunits UQCRC1 and UQCRC2 share homology with mitochondrial-processing peptidase (MPP) subunits, suggesting an additional role in the proteolytic processing of the Rieske protein (UQCRFS1) for complex III maturation.
- UQCRC1 localizes specifically to the mitochondrial inner membrane, as a fundamental membrane-embedded subunit of the respiratory chain complex.
- The protein is broadly expressed in metabolically active tissues, including brain (substantia nigra, striatum, cortex, cerebellum), spinal cord, heart, kidney, liver, and muscle.
- Post-translational modifications include acetylation and phosphorylation, identified through proteomics studies, which may regulate its function or complex assembly.
- Genetic variations in UQCRC1 have been linked to neurodegenerative diseases; the protein is associated with keywords such as Disease variant, Parkinsonism, and Neuropathy.
- UQCRC1 appears to play a critical role in maintaining proper mitochondrial function in nigral dopaminergic neurons, highlighting its potential relevance in Parkinson’s disease. Related references: PMID:33141179
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fragment covering amino acids 156–480 of human UQCRC1, expressed as a fusion protein. This region is well-conserved, so the antibody may recognize the target in its native conformation.
- For Western blotting, the predicted molecular weight of human UQCRC1 is approximately 53 kDa (UniProt annotation: 52.6 kDa). Band size may vary slightly across species or due to post-translational modifications.
- When performing IHC-P, standard antigen retrieval methods (e.g., heat-induced epitope retrieval) are recommended, as the epitope may be masked in formalin-fixed paraffin-embedded tissues.
- The antibody is validated for human, mouse, and rat samples. For other species, cross-reactivity should be experimentally confirmed.
- Suitable for multiple immunodetection techniques (WB, IHC-P, IF/ICC, IP, ELISA), but optimal working dilutions should be determined empirically for each application and sample type.
