Biological Background: UPF3A Function and Localization

• UPF3A (Regulator of nonsense transcripts 3A, also known as UPF3, HUPF3A, RENT3A) is a component of the nonsense-mediated mRNA decay (NMD) pathway. It associates with the exon junction complex (EJC) and bridges the EJC core to the NMD machinery, recruiting UPF2 at the cytoplasmic side of the nuclear envelope.
• Compared to its paralog UPF3B, UPF3A is only marginally active in NMD; however, it still binds spliced mRNA upstream of exon-exon junctions and weakly stimulates translation in vitro.
• The protein localizes to both the nucleus and cytoplasm, consistent with its role in mRNA surveillance and nucleocytoplasmic transport.
• Isoform 1 is strongly expressed in testis, uterus, muscle, fetal brain, and spinal cord. Isoform 2 is strongly expressed in fetal brain and spinal cord.
• UPF3A is a phosphoprotein with RNA-binding activity; it is implicated in alternative splicing and mRNA transport.
• The canonical isoform has a molecular weight of approximately 54.7 kDa; the immunogen-based antibody detects a band around 55 kDa.

• The immunogen spans amino acids 327–476 of human UPF3A (NP_075387.1). This region may be conserved across species, contributing to the observed cross-reactivity with mouse and rat.
• For Western blot, a predominant band at ~55 kDa is expected. Consider using lysates from testis, uterus, or muscle as positive controls based on reported tissue specificity.
• In immunofluorescence/ICC, UPF3A displays both nuclear and cytoplasmic staining; optimize fixation and permeabilization to preserve both localizations.
• As a polyclonal antibody, it may recognize multiple isoforms or degradation products. Validate specificity in the relevant sample system.
