- UPF2 (Regulator of nonsense transcripts 2), also known as up-frameshift suppressor 2 homolog (hUpf2), is encoded by the UPF2 gene (KIAA1408, RENT2) and is a core component of the nonsense-mediated mRNA decay (NMD) pathway.
- UPF2 is recruited to the exon junction complex (EJC) via UPF3B at the cytoplasmic side of the nuclear envelope, leading to the assembly of the UPF1–UPF2–UPF3 surveillance complex that triggers NMD of mRNAs containing premature stop codons.
- In cooperation with UPF3B, UPF2 stimulates both ATPase and RNA helicase activities of UPF1, thereby facilitating the degradation of aberrant transcripts.
