Biological Background: UPF1/RENT1 Function and Localization

UPF1 (Regulator of nonsense transcripts 1), also known as RENT1, NORF1, hUpf1, is an ATP-dependent RNA helicase essential for nonsense-mediated mRNA decay (NMD), a surveillance pathway that degrades aberrant mRNAs with premature stop codons and also modulates expression of normal mRNAs.

The protein is recruited to translating ribosomes upon termination, where it forms the transient SURF (SMG1-UPF1-eRF1-eRF3) complex. In EJC-dependent NMD, this complex associates with the exon junction complex via UPF2, forming the UPF1-UPF2-UPF3 surveillance complex that triggers mRNA degradation.
