Biological Background: UPF1/RENT1 Function and Localization

  • UPF1 (Regulator of nonsense transcripts 1), also known as RENT1, NORF1, hUpf1, is an ATP-dependent RNA helicase essential for nonsense-mediated mRNA decay (NMD), a surveillance pathway that degrades aberrant mRNAs with premature stop codons and also modulates expression of normal mRNAs.

  • The protein is recruited to translating ribosomes upon termination, where it forms the transient SURF (SMG1-UPF1-eRF1-eRF3) complex. In EJC-dependent NMD, this complex associates with the exon junction complex via UPF2, forming the UPF1-UPF2-UPF3 surveillance complex that triggers mRNA degradation.
