Biological Background: UNC5B Function and Localization
- Protein identity: Netrin receptor UNC5B (also known as protein unc-5 homolog 2, protein unc-5 homolog B, or p53-regulated receptor for death and life protein 1/p53RDL1) is a single-pass transmembrane receptor that belongs to the UNC5 netrin receptor family.
- Core function in axon guidance: UNC5B functions as a receptor for netrin, mediating axon repulsion of neuronal growth cones during development of the nervous system. This repulsive signaling may occur through association with DCC.
- Role in angiogenesis: Negatively regulates vascular branching by mediating retraction of tip cell filopodia on endothelial growth cones in response to netrin, thereby controlling blood vessel formation.
- Dependence receptor activity: Acts as a dependence receptor that triggers apoptosis in the absence of netrin ligand. UNC5B activates DAPK1 by reducing its autoinhibitory phosphorylation at Ser-308, increasing catalytic activity and promoting cell death. Related references: PMID:12598906
- Subcellular localization: Localizes to the cell membrane and is enriched in membrane rafts, consistent with its role in signal transduction at the cell surface.
- Post‑translational modifications: UNC5B is subject to alternative splicing, disulfide bond formation, glycosylation, palmitoylation, and phosphorylation, which may regulate its stability, localization, and activity.
- Tissue distribution: Highly expressed in brain; also detected at lower levels in developing lung, cartilage, kidney, and hematopoietic/immune tissues.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning amino acids 290–377 within the extracellular domain of human UNC5B. This region may be well‑exposed in native conformations, so consider its utility for detecting the full‑length protein (~104 kDa) in WB.
- For ELISA applications, the recombinant immunogen itself or purified UNC5B protein can serve as a positive control to validate assay sensitivity and specificity.
- Cross‑reactivity is reported for human, mouse, and rat; however, sequence variations between species within the immunogen region should be verified, and detection in each species should be empirically validated in the relevant sample system.
- Because UNC5B is a transmembrane receptor enriched in membrane rafts, adequate membrane solubilization (e.g., using non‑denaturing detergents) may improve detection efficiency.
