Biological Background: Uromodulin Function and Localization
- Uromodulin (UMOD; Tamm-Horsfall glycoprotein, THP) is the most abundant protein in normal human urine and is exclusively synthesized by epithelial cells of the thick ascending limb of Henle's loop (TALH) and distal convoluted tubule in the kidney.
- The protein localizes to the apical membrane, basolateral membrane, and primary cilium, and is secreted into the urine; it promotes apical membrane organization and forms a filamentous gel-like structure thought to regulate water barrier permeability.
- Uromodulin can act as a receptor for cytokines (IL-1, IL-2, TNF) and facilitate their endocytosis (PubMed:3498215), and it enhances neutrophil migration across renal epithelia (PubMed:20798515). Related references: PMID:3498215 PMID:20798515
- In urine, it contributes to colloid osmotic pressure, retards passage of positively charged electrolytes, and inhibits formation of supersaturated salt crystals (By similarity).
- Uromodulin protects against urinary tract infections by binding type 1 fimbriated E. coli via the bacterial adhesin fimH, preventing adherence to urothelial receptors and enabling pathogen clearance during micturation (PubMed:11134021, PubMed:32616672); it also aggregates other uropathogens like K. pneumoniae, P. aeruginosa, and S. mitis (PubMed:32616672). - The protein contains EGF-like domains, multiple disulfide bonds, and a C-terminal GPI-anchor; alternative splicing generates isoforms, and mutations in UMOD cause Mendelian kidney diseases such as medullary cystic kidney disease type 2 (MCKD2) and familial juvenile hyperuricemic nephropathy (FJHN).
- Predicted molecular weight is approximately 70 kDa (based on amino acid sequence), consistent with Western blot detection at ~70 kDa under reducing conditions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 315–615 of human UMOD, covering a region with multiple EGF-like domains; therefore, the antibody may recognize multiple isoforms and both membrane-associated and secreted forms.
- For Western blotting, UMOD typically migrates at ~70 kDa under reducing conditions; consider using human kidney lysate or urine as positive controls, and validate in the relevant sample system.
- For ELISA applications, optimize coating with recombinant protein or native urinary uromodulin; the antibody’s reactivity with mouse UMOD enables cross-species detection in murine models.
- Given uromodulin’s role in renal physiology and UTI defense, this antibody may be useful for studying expression in kidney diseases, bacterial adhesion assays, or cytokine binding studies; however, experimental conditions must be empirically determined.
