Biological Background: ULK1 Function and Localization
- Serine/threonine-protein kinase ULK1, the mammalian homolog of yeast ATG1, is a central initiator of autophagy. It acts upstream of the class III PI3K complex (PIK3C3) to regulate autophagophore formation and participates in regulatory feedback loops with mTORC1 and AMPK. - Phosphorylates multiple autophagy-related substrates, including ATG13, RPTOR, SESN2, SQSTM1/p62, FLCN, AMBRA1, and ATG4B, thereby coordinating autophagosome initiation, nucleation, and maturation.
