Biological Background: ULBP2 Function and Localization
- UL16-binding protein 2 (ULBP2), also known as ALCAN-alpha, N2DL2, NKG2DL2, or RAET1H, is a member of the ULBP/RAET1 family of MHC class I-related molecules.
- ULBP2 functions as a ligand for the KLRK1/NKG2D receptor, an activating receptor expressed on natural killer (NK) cells, CD8+ T cells, and γδ T cells.
- Binding of ULBP2 to NKG2D triggers NK cell-mediated cytotoxicity, playing a critical role in immune surveillance against tumors and infected cells.
- ULBP2 is a GPI-anchored glycoprotein expressed on the cell membrane; it can also be found in the endoplasmic reticulum and as a secreted form.
- The protein contains a signal peptide, an extracellular region with MHC class I-like domains, and a C-terminal GPI-anchor attachment site; it has a predicted molecular weight of approximately 27 kDa.
- ULBP2 is preferentially expressed in various cancer cell lines and in fetal tissues, but is generally absent in normal adult tissues, making it an attractive target for cancer immunotherapy.
