Biological Background: ULBP1 Function and Localization
- ULBP1 (UL16-binding protein 1), also known as N2DL-1, RAET1I, or NKG2DL1, is a GPI-anchored cell surface glycoprotein and a member of the RAET1 family.
- It functions as a stress-induced ligand for the activating receptor NKG2D (KLRK1), playing a critical role in immune surveillance by mediating natural killer (NK) cell cytotoxicity.
- ULBP1 is localized to the cell membrane and endoplasmic reticulum, and is widely expressed in tissues including heart, brain, lung, liver, testis, lymph node, thymus, tonsil, bone marrow, and also in T-cells, B-cells, and erythroleukemia cell lines.
- The protein contains a signal peptide, an extracellular domain with disulfide bonds, and a C-terminal GPI-anchor for membrane attachment.
- It is implicated in host-virus interactions and may be involved in the recognition and elimination of infected or transformed cells by the innate immune system.
- UniProt keywords include: Cell membrane, Disulfide bond, Endoplasmic reticulum, Glycoprotein, GPI-anchor, Host-virus interaction, Immunity, Lipoprotein, Membrane, Signal.
- Its molecular weight is approximately 28 kDa (calculated 27997 Da from UniProt, observed 28kDa in product specifications).
Experimental Guidance and Technical Tips
- For Western blot, the predicted band size is ~28 kDa; consider using appropriate positive controls such as human or mouse tissues expressing ULBP1.
- For immunohistochemistry, antigen retrieval may be required; validate in tissue sections known to express ULBP1, such as lymph node or testis.
- This antibody was generated using a recombinant protein immunogen (aa 26-216), so it recognizes an epitope within the extracellular domain; it may detect both membrane-bound and soluble forms.
- Due to cross-reactivity with mouse, it can be used in mouse models to study NKG2D-mediated immunity; however, verify specificity in your system.
- ELISA can be employed for quantitative detection; titration and optimization are recommended.
