  • ULBP1 (UL16-binding protein 1), also known as ALCAN-beta, NKG2D ligand 1 (N2DL-1/NKG2DL1), and retinoic acid early transcript 1I, is a cell-surface glycoprotein belonging to the RAET1 family. It shares structural similarity with MHC class I molecules but does not present peptides.
  • The primary function of ULBP1 is to bind and activate the KLRK1/NKG2D receptor on natural killer (NK) cells, thereby triggering NK cell-mediated cytotoxicity and cytokine release, crucial for immune surveillance against infected or transformed cells.
  • ULBP1 is a GPI-anchored membrane protein localized mainly at the cell membrane, but it is also detected in the endoplasmic reticulum, reflecting its secretory pathway processing and regulation.

  • The immunogen consists of a recombinant fusion protein corresponding to the N-terminal region (amino acids 1–100) of human ULBP1 (NP_079494.1). This fragment covers the signal peptide and part of the extracellular domain, which may be suitable for detecting both full-length endogenous and recombinant ULBP1.
  • For western blot (WB) applications, consider using whole-cell lysates from ULBP1-expressing cell lines such as K562 (erythroleukemia) or Jurkat (T-cell leukemia). Given the predicted molecular weight of ~28 kDa (core protein; glycosylation may increase apparent mass), optimize reducing conditions and protein loading.
  • The antibody also cross-reacts with mouse ULBP1, making it a potential tool for cross-species studies. However, validate reactivity in the relevant mouse tissue or cell models, as epitope conservation may affect performance.
  • In ELISA, consider pairing this monoclonal antibody with a polyclonal detection antibody to establish a sandwich assay for soluble ULBP1, which can be shed from the cell surface. Optimize the coating and detection steps for sensitivity.
