- RAP80 is a ubiquitin-binding protein that specifically recognizes and binds Lys-63-linked ubiquitin chains with high affinity, while showing weak binding to monoubiquitin and no affinity for Lys-48-linked chains. Related references: PMID:24627472, PMID:19328070
- Core component of the BRCA1-A complex, directing BRCA1-BARD1 to DNA double-strand breaks via binding to Lys-63-ubiquitinated histones H2A and H2AX.
- The BRCA1-A complex also possesses deubiquitinase activity that specifically removes Lys-63-linked ubiquitin from histones H2A and H2AX at damage sites.
- May function as a transcriptional repressor by inhibiting NR6A1 interaction with NCOR1 corepressor.
- Localizes predominantly to the nucleus, consistent with its role in DNA damage signaling and chromatin regulation.
- Expressed in testis, ovary, thymus, and heart; also detected in testicular germ cells.
- Post-translational modifications include phosphorylation and ubiquitination; contains zinc-finger domains indicative of DNA repair function.
