Biological Background: UGT1A6 Function and Localization
- UGT1A6 is a UDP-glucuronosyltransferase that catalyzes phase II biotransformation, conjugating lipophilic substrates with glucuronic acid for elimination and detoxification of drugs, xenobiotics, and endogenous compounds. Related references: PMID:15231852 PMID:21422672
- It plays a role in the glucuronidation of arachidonic acid and its eicosanoid derivatives, such as 15-HETE and 20-HETE. Related references: PMID:15231852
- The enzyme preferentially conjugates small planar phenolic molecules (e.g., 4-nitrophenol, 1-naphthol) but not bulky phenols, androgens, or estrogens. 2-hydroxybiphenyl is an excellent substrate.
- UGT1A6 is involved in glucuronidation of the phytochemical ferulic acid at the phenolic or carboxylic acid group. Related references: PMID:21422672
- Alternative splicing produces isoform 3, which lacks transferase activity but acts as a negative regulator of isoform 1.
- Subcellularly, UGT1A6 localizes to the microsome and endoplasmic reticulum membrane, consistent with its role in detoxification pathways.
- Tissue expression includes skin; isoforms 1 and 3 are found in kidney and liver; isoform 1 is also detected in colon, esophagus, and small intestine.
- Post-translational modifications include glycosylation, and it contains a signal sequence and transmembrane helix, as indicated by UniProt keywords.
Experimental Guidance and Technical Tips
- For Western blotting, consider using a suitable loading control and validate band specificity with knockdown or knockout controls.
- Given the immunogen design covering the core glycosyltransferase domain, the antibody may recognize multiple UGT1A isoforms sharing high sequence homology; verify specificity in your experimental system.
- For immunofluorescence, permeabilization may be required to access the ER membrane, and include appropriate negative controls to confirm signal specificity.
- In ELISA applications, optimize coating conditions and antibody dilutions using relevant recombinant protein standards.
