Biological Background: UGT1A6 Function and Localization
- UGT1A6 is a member of the UGT1 family and functions as a phenol-metabolizing UDP-glucuronosyltransferase (UDPGT) critical for the detoxification of drugs, xenobiotics, and endogenous compounds by conjugating them with glucuronic acid. Related references: PMID:15231852, PMID:21422672
- Exhibits broad substrate specificity towards small planar phenols, including 4-nitrophenol, 1-naphthol, and 4-methylumbelliferone, but does not conjugate bulky phenols or steroid hormones.
- Involved in the glucuronidation of the dietary phytochemical ferulic acid, utilizing both phenolic and carboxylic acid groups. Related references: PMID:21422672
- Plays a role in the glucuronidation of arachidonic acid and its derivatives 15-HETE and 20-HETE, linking it to eicosanoid metabolism.
- Isoform 3, generated by alternative splicing, lacks transferase activity and acts as a negative regulator of isoform 1.
- Primarily localized in the membrane of the endoplasmic reticulum and microsomes.
- Tissue expression includes liver, kidney, skin, colon, esophagus, and small intestine, with isoform-dependent distribution.
- Contains N-glycosylation sites and a transmembrane domain, consistent with its membrane-bound transferase function.
Experimental Guidance and Technical Tips
- For Western blot, expect a specific band around 61 kDa (predicted molecular weight). Use reducing conditions and validate with appropriate positive controls (e.g., human liver microsomes or HepG2 cell lysates).
- Since polyclonal antibodies may recognize multiple isoforms, consider confirming isoform specificity by consulting sequence alignment and testing in isoform-expressing systems.
- For ELISA, the immunogen corresponds to amino acids 65-270; ensure the coating antigen includes this region. Perform checkerboard titrations to optimize antibody and antigen concentrations.
- Cross-reactivity has been detected with human and mouse; if extending to other species, validate sequence homology of the immunogen region.
