Biological Background: UFM1 Function and Localization
- UFM1 (Ubiquitin-fold modifier 1) is a 9 kDa ubiquitin-like protein encoded by the UFM1 gene (formerly C13orf20). It is also known as BM-002 and is associated with leukodystrophy (HLD14) through alternative splicing and disease variants.
- UFM1 is covalently attached to lysine residues of substrate proteins via an enzymatic cascade involving the E1 enzyme UBA5, E2 enzyme UFC1, and E3 ligase UFL1, in a process termed ufmylation. Related references: PMID:15071506 PMID:20018847 PMID:27653677
- Ufmylation participates in ribosome recycling, response to DNA damage, transcription, and reticulophagy (ER-phagy) triggered by endoplasmic reticulum stress. Related references: PMID:25219498 PMID:32160526 PMID:38383785
- Subcellularly, UFM1 localizes to both the nucleus and cytoplasm, consistent with its involvement in nuclear and cytoplasmic processes.
- The protein features an isopeptide bond essential for conjugation, and its three-dimensional structure has been resolved (3D-structure). It is subject to alternative splicing and bears disease variants linked to leukodystrophy.
Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against the N-terminal region (aa 1-63) of human UFM1. Given high cross-species conservation, it is expected to recognize human, mouse, and rat UFM1 as indicated.
- For Western blot (WB), use a total protein loading control and optimize transfer conditions for low-molecular-weight proteins (~9 kDa).
- In immunoprecipitation (IP) or proximity ligation assays, the antibody detects total UFM1 but may not distinguish free versus conjugated forms without further validation.
- For IHC-P and IF/ICC, determine appropriate antigen retrieval and permeabilization protocols, as UFM1 is present in both nucleus and cytoplasm.
- This antibody is suitable for ELISA applications; pairing with a specific detection antibody may require optimization.
