Biological Background: UFD1L Function and Localization
- UFD1L (Ubiquitin fusion degradation protein 1-like), also known as Ub fusion protein 1, is an essential component of the ubiquitin-dependent proteolytic pathway that degrades ubiquitin fusion proteins.
- It forms a ternary complex with VCP/p97 and NPLOC4, which recognizes ubiquitinated substrates and facilitates the retrotranslocation and export of misfolded proteins from the endoplasmic reticulum (ER) to the cytoplasm for proteasomal degradation.
- The UFD1-VCP-NPLOC4 complex also plays a critical role in mitotic spindle disassembly at the end of mitosis and is required for closed nuclear envelope formation.
- UFD1L negatively regulates type I interferon production by forming a complex with VCP and NPLOC4 that binds to RIG-I, recruiting the E3 ubiquitin ligase RNF125 to promote ubiquitination and subsequent degradation of RIG-I. Related references: PMID:26471729
- Subcellularly, UFD1L is found in the nucleus, cytoplasm, and predominantly in the cytosol.
- Tissue expression includes adult heart, skeletal muscle, and pancreas, as well as fetal liver and kidney.
- Post-translational modifications include acetylation and phosphorylation; the UFD1 gene undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen region (aa 1-307) covers a large N-terminal portion of UFD1L, which may enable detection of multiple isoforms; however, validate specificity in the relevant sample system.
- For Western blot analysis, expect a band near the predicted molecular weight of 35 kDa (34.5 kDa theoretical). Use appropriate positive controls (e.g., HeLa, HEK293).
- In immunofluorescence/ICC, optimize fixation and permeabilization conditions, as UFD1L localizes to both nuclear and cytoplasmic compartments.
- For ELISA applications, use the recombinant immunogen or purified protein as a positive control.
