Biological Background: UFD1L Function and Localization
- Encoded by the UFD1L gene, ubiquitin fusion degradation protein 1 (UFD1) is a 307-amino acid protein also known as UB fusion protein 1, with a predicted molecular weight of ~35 kDa.
- Essential component of the ubiquitin-dependent proteolytic pathway; degrades ubiquitin fusion proteins and mediates export of misfolded proteins from the ER to the cytoplasm for proteasomal degradation as part of the UFD1-VCP-NPLOC4 complex.
- The UFD1-VCP-NPLOC4 complex regulates spindle disassembly at the end of mitosis and is necessary for the formation of a closed nuclear envelope. May be involved in the development of some ectoderm-derived structures (By similarity).
- Acts as a negative regulator of type I interferon production; the UFD1-VCP-NPLOC4 complex binds to RIGI and recruits RNF125, promoting ubiquitination and degradation of RIGI. Related references: PMID:26471729
- Found predominantly in the nucleus and cytoplasm/cytosol.
- Tissue specific expression: adult heart, skeletal muscle, pancreas, and fetal liver and kidney.
- Post-translationally modified by acetylation and phosphorylation; alternative splicing yields multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to full-length human UFD1L (aa 1-307), so the antibody may recognize multiple isoforms. Consider verifying isoform detection in relevant cell lysates.
- For Western Blot, expect a band at ~35 kDa; validate using appropriate positive controls such as HeLa or HEK293 cell lysates.
- For ELISA, use recombinant UFD1L protein as the coating antigen for direct ELISA. Sandwich ELISA may require a matched antibody pair; test pairing with monoclonal antibodies targeting distinct epitopes.
- Cross-reactivity with mouse and rat is reported; confirm in endogenous tissue or cell line samples from these species before quantitative applications.
- Optimize antibody dilution and incubation conditions empirically for each assay and sample type.
