Biological Background: UCP2 Function and Localization
- UCP2, encoded by the UCP2 gene (GeneID: 7351), is a mitochondrial carrier protein that functions as an antiporter exporting dicarboxylate Krebs cycle intermediates (e.g., oxaloacetate) in exchange for phosphate and a proton, thereby downregulating acetyl-CoA oxidation and reducing reactive oxygen species production. Related references: PMID:24395786
- It may mediate inducible proton entry into the mitochondrial matrix, affecting ATP turnover and protecting against oxidative stress, a process likely associated with fatty acid flipping across the inner membrane. Related references: PMID:11171965, PMID:11278935, PMID:22524567
- UCP2 can unidirectionally transport anions including L-malate, L-aspartate, phosphate, and chloride ions. Related references: PMID:22524567, PMID:24395786, PMID:26182433
- It is required for glucose-induced DRP1-dependent mitochondrial fission and neuronal activation in the ventromedial hypothalamus, contributing to systemic glucose homeostasis.
- During the resolution of inflammation, UCP2 attenuates ROS production in response to IL33, preserving Krebs cycle integrity for itaconate production and promoting differentiation of alternatively activated macrophages.
- UCP2 localizes to the mitochondrial inner membrane and is widely expressed in adult human tissues, with highest levels in white adipose tissue and skeletal muscle.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 112–211 of human UCP2, a region that may be conserved across species, potentially contributing to the observed cross-reactivity with mouse and rat.
- For Western blot, a band around 33 kDa is expected; validate using appropriate controls and reference lysates from target tissues.
- ELISA applications may require optimization of coating conditions and antibody dilution; consider testing with recombinant UCP2 protein or peptide.
