Biological Background: UCP1 Function and Localization
- Mitochondrial brown fat uncoupling protein 1 (UCP1), also known as Solute carrier family 25 member 7 (Slc25a7) or Thermogenin, is encoded by the Ucp1 gene in mice.
- Functions as a long-chain fatty acid (LCFA)/proton symporter, simultaneously transporting one LCFA and one proton across the inner mitochondrial membrane, dissipating the proton gradient and converting oxidative energy into heat instead of ATP. Related references: PMID:23063128
- Plays a central role in non-shivering adaptive thermogenesis within brown adipose tissue and beige fat, responding to cold exposure and diet to regulate whole-body energy expenditure. Related references: PMID:19187776 PMID:23063128 PMID:27027295
- Modulates mitochondrial reactive oxygen species (ROS) production, thereby influencing cellular redox homeostasis. Related references: PMID:20416274 PMID:20466728
- Localizes to the mitochondrial inner membrane and contains six transmembrane helices, characteristic of the mitochondrial solute carrier family.
- Expression is predominantly restricted to brown adipose tissue, consistent with its specialized role in thermogenesis.
- Classified as a selenoprotein; incorporates selenocysteine and belongs to the mitochondrial transporter family with ion channel/transport activities.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide derived from mouse UCP1, ensuring target specificity; verify the peptide sequence if cross-reactivity is critical for your model.
- For Western blot (WB), a band at ~33 kDa is expected in mouse and rat tissue lysates; use appropriate positive controls such as brown adipose tissue.
- For immunohistochemistry (IHC-P), antigen retrieval optimization may be necessary due to the protein's membrane-embedded conformation; consider heat-mediated or enzymatic methods.
- In immunoprecipitation (IP) and ELISA, validate the antibody under your specific buffer and detection conditions to confirm performance.
