Biological Background: UCHL3 Function and Localization
- UCHL3 (Ubiquitin carboxyl-terminal hydrolase isozyme L3), also known as ubiquitin thioesterase L3, is encoded by the UCHL3 gene in humans.
- UCHL3 is a thiol protease deubiquitinating enzyme (DUB) that hydrolyzes the C-terminal glycine of ubiquitin or NEDD8, controlling cellular ubiquitin pools through processing of ubiquitin precursors and ubiquitinated proteins. Related references: PMID:19154770 PMID:21762696 PMID:22689415
- The enzyme exhibits a 10-fold preference for Arg/Lys at position P3'' and preferentially cleaves 'Lys-48'-linked ubiquitin chains. Related references: PMID:19154770 PMID:21762696 PMID:22689415
- UCHL3 promotes DNA repair via homologous recombination by deubiquitinating RAD51 in response to DNA damage, thereby facilitating RAD51–BRCA2 interaction. Related references: PMID:27941124
- Additional functions include processing the neurodegeneration-associated UBB(+1) mutant ubiquitin, regulating ENaC-mediated apical membrane recycling, and enhancing insulin signaling via IGF1R/AKT/FOXO1 phosphorylation (By similarity).
- Highly expressed in heart, skeletal muscle, and testis; primarily localized in the cytoplasm.
- Classified as a phosphoprotein and member of the ubiquitin carboxyl-terminal hydrolase family, highlighting its role in the Ubl conjugation pathway.
Experimental Guidance and Technical Tips
- The immunogen spans the full-length human UCHL3 protein (amino acids 1–220); in Western blotting, expect a specific band near 26 kDa under reducing conditions. Validate detection in your target sample type.
- Reported cross‑reactivity with mouse and rat UCHL3; however, we recommend confirming reactivity with your specific model systems before large‑scale experiments.
- For ELISA applications, the antibody can be used as a capture or detection reagent; optimal working concentrations and buffer conditions should be empirically determined.
- Due to UCHL3’s cytoplasmic localization, use cytosolic fractions or mild lysis buffers to ensure efficient extraction of the native protein.
