Biological Background: UBQLN1 Function and Localization

  • Ubiquilin-1 is a 62.5 kDa protein encoded by the UBQLN1 gene, containing an N-terminal ubiquitin-like (UBL) domain and a C-terminal ubiquitin-associated (UBA) domain. These domains mediate interactions with proteasomal subunits and polyubiquitinated substrates, respectively, to facilitate proteasomal degradation (PubMed:15147878). Related references: PMID:15147878
  • It participates in endoplasmic reticulum-associated protein degradation (ERAD) by forming a link between polyubiquitinated ERAD substrates and the proteasome through interactions with ER-localized proteins UBXN4, VCP, and HERPUD1 (PubMed:18307982, PubMed:19822669).   • For Western blotting, the predicted molecular weight is approximately 63 kDa. Use appropriate blocking and wash conditions, and validate with positive and negative controls.
