Biological Background: UBL4A Function and Localization
- Ubiquitin-like protein 4A (UBL4A), also known as GDX, GET5, MDY2, TMA24, and DX254E, is a component of the BAG6/BAT3 protein quality control complex.
- As part of the BAG6/BAT3 complex, UBL4A maintains misfolded and hydrophobic patches-containing proteins in a soluble state and facilitates their delivery to the endoplasmic reticulum or proteasome (PubMed:20676083, PubMed:21636303, PubMed:21743475). Related references: PMID:20676083 PMID:21636303 PMID:21743475
- The complex mediates post-translational delivery of tail-anchored/type II transmembrane proteins to the ER membrane, interacting with the nascent transmembrane region at the ribosome (PubMed:20676083, PubMed:25535373, PubMed:28104892). Related references: PMID:20676083 PMID:25535373 PMID:28104892
- UBL4A is involved in sorting mislocalized secretory pathway proteins, either targeting them to the proteasome for degradation or redirecting them to the ER (PubMed:21743475). Related references: PMID:21743475
- It contributes to ER-associated degradation (ERAD) by maintaining retrotranslocated ER proteins in an unfolded, soluble state for proteasomal delivery (PubMed:21636303). Related references: PMID:21636303
- UBL4A localizes to the cytoplasm, cytosol, and nucleus.
- It is annotated as a phosphoprotein and can undergo ubiquitin-like conjugation; it is also associated with an isopeptide bond.
Experimental Guidance and Technical Tips
- The immunogen covers the full-length protein (aa 1-157), enabling detection of both endogenous UBL4A and overexpressed constructs.
- For Western blot, a band near 18 kDa is expected; validate specificity using human, mouse, or rat lysates and consider loading controls.
- Polyclonal antibodies may require titration to minimize non-specific background; optimization of blocking and primary antibody dilution is advised.
- In immunoprecipitation, be aware that UBL4A is part of a dynamic complex (BAG6/BAT3) and adjust lysis/wash conditions to preserve native interactions or denature as needed.
- ELISA protocols should be optimized for coating antigen and antibody concentration, as the antibody is raised against recombinant protein but not pre-validated for specific sandwich pairs.
