- For Western blotting (WB), a band around 26 kDa may correspond to polyubiquitin-B precursor, but individual ubiquitin monomers (~8.5 kDa) conjugated to targets will appear as higher molecular weight smears. Optimize sample preparation and detection conditions to resolve polyubiquitinated species.
- In IHC-P and IF/ICC, verify specificity by comparing staining patterns in known positive and negative tissues/cell lines, and include a blocking peptide competition assay if available.
- For ELISA applications, ensure that the recombinant immunogen or purified ubiquitin is used as a standard to validate sensitivity and linearity in your assay system.
