Biological Background: Ubiquilin-2 Function and Localization

  • Ubiquilin-2 (UBQLN2), also known as DSK2 homolog, PLIC-2, CHAP1, and N4BP4, is a ubiquitin-like protein encoded by the UBQLN2 gene.
  • It plays an important role in the regulation of different protein degradation mechanisms and pathways including ubiquitin-proteasome system (UPS), autophagy, and endoplasmic reticulum-associated protein degradation (ERAD). Mediates the proteasomal targeting of misfolded or accumulated proteins for degradation by binding (via UBA domain) to their polyubiquitin chains and by interacting (via ubiquitin-like domain) with the subunits of the proteasome (PubMed:10983987). Related references: PMID:10983987
  • UBQLN2 participates in the ERAD pathway via its interaction with ER-localized proteins FAF2/UBXD8 and HERPUD1 and may form a link between the polyubiquitinated ERAD substrates and the proteasome (PubMed:18307982, PubMed:24215460). Related references: PMID:18307982, PMID:24215460
  • Involved in the regulation of macroautophagy and autophagosome formation; required for maturation of autophagy-related protein LC3 from the cytosolic form LC3-I to the membrane-bound form LC3-II and may assist in the maturation of autophagosomes to autolysosomes by mediating autophagosome-lysosome fusion (PubMed:19148225, PubMed:20529957). Related references: PMID:19148225, PMID:20529957
  • Negatively regulates the endocytosis of GPCR receptors: AVPR2 and ADRB2, by specifically reducing the rate at which receptor-arrestin complexes concentrate in clathrin-coated pits (CCPs) (PubMed:18199683). Related references: PMID:18199683
  • Subcellularly, UBQLN2 localizes to the cytoplasm, nucleus, membrane, and cytoplasmic vesicles including autophagosomes.
  • Post-translational modifications include acetylation; mutations in UBQLN2 are associated with amyotrophic lateral sclerosis (ALS) and neurodegeneration.

Experimental Guidance and Technical Tips

  • The immunogen encompasses the N-terminal 1-150 amino acids of human UBQLN2; consider epitope accessibility and potential isoform-specific recognition in your assay design.
  • For Western blot, validate antibody specificity using relevant positive controls (e.g., human or mouse cell lysates) and consider siRNA knockdown or CRISPR knock-out controls to confirm target band identity.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and permeabilization protocols; a nuclear/cytoplasmic staining pattern is expected.
  • This antibody cross-reacts with mouse, allowing its use in mouse tissue or cell models; verify cross-reactivity in your specific system.
  • As a rabbit polyclonal, it is compatible with standard anti-rabbit secondary detection reagents for colorimetric, fluorescent, or ELISA applications.
