Biological Background: UBE2V1 Function and Localization

  • UBE2V1, also known as CROC-1 or TRAF6-regulated IKK activator 1 beta Uev1A, is a ubiquitin-conjugating enzyme E2 variant that lacks intrinsic ligase activity but functions as a cofactor with UBE2N.
  • Forms a heterodimer with UBE2N to catalyze the synthesis of non-canonical Lys-63-linked polyubiquitin chains, which play critical roles in signal transduction rather than proteasomal degradation.
  • The UBE2V1-UBE2N complex mediates Lys-63 polyubiquitination events that activate NF-kB signaling downstream of IL1B, TNF, TRAF6, and TRAF2, promoting inflammatory and immune responses.
  • Involved in error-free DNA repair and contributes to cell survival after DNA damage, as well as controlling cell cycle progression and differentiation.
  • Together with RNF135 and UBE2N, catalyzes the viral RNA-dependent 'Lys-63'-linked polyubiquitination of RIGI to activate the downstream signaling pathway that leads to interferon beta production. Related references: PMID:31006531
