Biological Background: UBE2S Function and Localization
- UBE2S, also known as E2-EPF, is a ubiquitin-conjugating enzyme (E2) that plays a critical role in protein ubiquitination and cell cycle control.
- UBE2S accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to substrate proteins, mediating polyubiquitination (PubMed:19820702, PubMed:19822757, PubMed:22496338). Related references: PMID:19820702 PMID:19822757 PMID:22496338
- It specifically catalyzes 'Lys-11'-linked polyubiquitin chain formation and acts as an essential factor of the anaphase-promoting complex/cyclosome (APC/C), a ubiquitin ligase that controls mitotic progression (PubMed:19820702, PubMed:19822757, PubMed:27259151). Related references: PMID:19820702 PMID:19822757 PMID:27259151
- UBE2S elongates 'Lys-11'-linked polyubiquitin chains initiated by the E2 enzyme UBE2C on APC/C substrates, thereby promoting the degradation of these substrates by the proteasome and facilitating mitotic exit (PubMed:19820702, PubMed:19822757, PubMed:27259151). Related references: PMID:19820702 PMID:19822757 PMID:27259151
- In addition to its role in cell cycle control, UBE2S is involved in the ubiquitination and subsequent degradation of VHL, leading to the accumulation of HIF1A (PubMed:16819549). Related references: PMID:16819549
- In vitro, UBE2S can promote polyubiquitination using all seven ubiquitin Lys residues, with the exception of 'Lys-48'-linked chains (PubMed:20061386, PubMed:20622874). Related references: PMID:20061386 PMID:20622874
- UBE2S is subject to post-translational modifications, including phosphorylation and acetylation, which may regulate its activity and stability.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein corresponding to amino acids 122-221 of human UBE2S, covering part of the catalytic domain. This design may yield antibodies that recognize both native and denatured UBE2S.
- For Western blotting (WB), consider using whole-cell lysates from human, mouse, or rat cell lines, as the antibody has shown cross-reactivity with these species. A band at approximately 24 kDa is expected.
- For ELISA applications, validate the antibody with your specific antigen and assay conditions. The supplier has validated the antibody in ELISA, but individual optimization is recommended.
- The antibody is KO validated, indicating specific reactivity towards UBE2S. Nevertheless, it is advisable to perform appropriate controls in your own experimental settings.
