Biological Background: UBE2O Function and Localization
- UBE2O (Ubiquitin-conjugating enzyme E2 O) is also known as E2-230K or 2O, a large 141 kDa E2/E3 hybrid ubiquitin-protein ligase.
- It displays both E2 and E3 ligase activities and mediates monoubiquitination of diverse target proteins. Related references: PMID:23455153, PMID:24703950
- Acts as a positive regulator of BMP7 signaling by monoubiquitinating SMAD6, thereby regulating adipogenesis. Related references: PMID:23455153
- Negatively regulates TRAF6-mediated NF-κB activation independently of its E2 activity. Related references: PMID:23381138
- Monoubiquitinates the nuclear localization signal (NLS) of BAP1, causing cytoplasmic retention, and similarly regulates INO80 and CXXC1 localization. Related references: PMID:24703950
- Assists the TRIM27:MAGEL2 E3 ligase complex in mediating Lys-63-linked ubiquitination of WASHC1 to promote endosomal F-actin assembly and retrograde transport. Related references: PMID:23452853
- Monoubiquitinates cytoplasmic CTNNA1 to trigger a binding switch from β-catenin to vinculin, driving cell-to-extracellular matrix adhesion. Related references: PMID:40983751
- Found in both cytoplasm and nucleus; predominantly expressed in skeletal muscle and heart. Phosphorylated and contains ATP-binding and coiled-coil domains.
Experimental Guidance and Technical Tips
- The immunogen covers residues 1073–1292 near the C‑terminus; consider peptide competition to verify band identity in new sample systems.
- In WB, expect a dominant band at approximately 141 kDa. Use KO‑validated lysates as a negative control to confirm specificity.
- For IF/ICC, optimize permeabilization and fixation since UBE2O shuttles between cytoplasm and nucleus.
- For IP, confirm compatibility of the rabbit polyclonal antibody with protein A/G resins and validate elution conditions.
- ELISA may be used for titer determination or sandwich pair screening; pair with a detection antibody recognizing a distinct epitope.
