Biological Background: Ube2L3 / UBCH7 Function and Localization
- UBE2L3 is a ubiquitin-conjugating enzyme E2 that specifically partners with HECT-type and RBR family E3 ubiquitin ligases, including PRKN and RNF31. It lacks intrinsic reactivity with lysine for most RING-type E3s.
- It accepts ubiquitin from the E1 enzyme and catalyzes covalent attachment to target proteins, mediating ubiquitination via complexes such as CUL9-RBX1 (PubMed:38605244). Related references: PMID:38605244
- UBE2L3 catalyzes 'Lys-11'-linked polyubiquitination in vitro and plays a key role in the selective degradation of short-lived and abnormal proteins.
- The enzyme is down-regulated during S-phase and is involved in cell cycle progression, as well as in the regulation of nuclear hormone receptor transcriptional activity.
- It may also play a role in myelopoiesis.
- The protein is localized to both the nucleus and cytoplasm, and is ubiquitously expressed with highest levels in testis.
- UBE2L3 is subject to acetylation and alternative splicing, and is identified as a reference proteome component involved in transcription regulation and ubiquitin conjugation.
Experimental Guidance and Technical Tips
- The antibody was generated against a synthetic peptide spanning amino acids 55-154 of human UBE2L3. For applications, consider the reported molecular weight of approximately 18 kDa for target detection in Western blotting.
- Given its reactivity with human, mouse, and rat samples, this antibody may be suitable for cross-species studies; however, validation in each species is recommended.
- For immunohistochemistry (IHC) and immunofluorescence (IF/ICC), the nuclear and cytoplasmic localization of UBE2L3 suggests that permeabilization steps should be optimized to ensure detection in both compartments.
- For ELISA and other sensitive assays, antibody titration is advised to maximize signal-to-noise ratio.
