Biological Background: UBE2L3 Function and Localization

• Ubiquitin-conjugating enzyme E2 L3 (UBE2L3, also known as UbcH7, L-UBC, E2-F1) is a member of the ubiquitin-conjugating enzyme family that plays a central role in the ubiquitin-proteasome pathway.
• It specifically functions with HECT-type and RBR family E3 ubiquitin-protein ligases, but lacks intrinsic reactivity with most RING-containing E3 ligases; it shows activity with RBR hybrids like PRKN, RNF31, and ARIH1.
• UBE2L3 accepts ubiquitin from the E1 enzyme and catalyzes its covalent attachment to substrate proteins; in vitro it mediates 'Lys-11'-linked polyubiquitination.
