Biological Background: UBE2L3 Function and Localization
- Ubiquitin-conjugating enzyme E2 L3 (UBE2L3, also known as UbcH7, L-UBC, E2-F1) is a member of the ubiquitin-conjugating enzyme family that plays a central role in the ubiquitin-proteasome pathway.
- It specifically functions with HECT-type and RBR family E3 ubiquitin-protein ligases, but lacks intrinsic reactivity with most RING-containing E3 ligases; it shows activity with RBR hybrids like PRKN, RNF31, and ARIH1.
- UBE2L3 accepts ubiquitin from the E1 enzyme and catalyzes its covalent attachment to substrate proteins; in vitro it mediates 'Lys-11'-linked polyubiquitination.
- Mediates ubiquitination by the CUL9-RBX1 complex. Related references: PMID:38605244
- Involved in the selective degradation of short-lived and abnormal proteins, and its expression is down-regulated during S-phase, linking it to cell cycle progression.
- Regulates the transcriptional activity of nuclear hormone receptors and may play a role in myelopoiesis.
- Subcellularly localized to both the nucleus and cytoplasm, with ubiquitous tissue expression and highest levels in testis.
- Post-translational modifications include acetylation; also subject to alternative splicing.
