Biological Background: UBAC2 Function and Localization

  • UBAC2 (also known as PHGDHL1) is a ubiquitin-associated domain-containing protein that functions as an ER-phagy receptor, mediating selective autophagic degradation of endoplasmic reticulum (ER) fragments to maintain ER homeostasis and restrain ER stress-induced inflammatory responses. This function depends on phosphorylation-induced dimerization, which enhances interaction with GABARAP and facilitates recruitment of ER fragments to autophagosomes. Related references: PMID:39284914

  • UBAC2 restricts the trafficking of FAF2 from the endoplasmic reticulum to lipid droplets. Related references: PMID:23297223

  • In association with LMBR1L and the E3 ubiquitin-protein ligase AMFR, UBAC2 negatively regulates the canonical Wnt signaling pathway in lymphocytes by promoting ubiquitin-mediated degradation of CTNNB1 and Wnt receptors FZD6 and LRP6. Related references: PMID:31073040

  • The protein localizes to the endoplasmic reticulum membrane and is characterized by a single-pass transmembrane domain.

  • UniProt lists additional post-translational modifications including glycosylation and phosphorylation, suggesting regulatory mechanisms that may modulate its function.

  • UBAC2 has a molecular weight of approximately 39 kDa (UniProt: 38964 Da).

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 220–310 of human UBAC2, a region conserved across human, mouse, and rat, which aligns with the reported cross-reactivity.
  • For Western blot, the expected band is around 39 kDa. Consider using appropriate positive controls (e.g., cell lines expressing UBAC2) and validating specificity with siRNA knockdown or knockout lysates.
  • In immunofluorescence/ICC, UBAC2 displays an ER membrane pattern. Optimize fixation and permeabilization conditions to preserve membrane staining and reduce background.
  • For ELISA, the antibody can be used as a detection or capture reagent; validate performance with recombinant UBAC2 protein or cell lysates to determine optimal working concentrations.
  • Given the target’s role in autophagy and Wnt signaling, functional assays may require careful consideration of cellular context and stimulation conditions.
