Biological Background: UBA52 (Ubiquitin-Ribosomal Protein eL40 Fusion Protein) Function and Localization
- UBA52 encodes a ubiquitin-ribosomal protein eL40 fusion protein, also known as CEP52, RPL40, or HUBCEP52. The precursor is processed to release free ubiquitin and ribosomal protein L40.
- The ubiquitin moiety can form isopeptide linkages with target proteins as monoubiquitin or polyubiquitin chains linked through Lys-6, -11, -29, -33, -48, -63, or linear Met-1, directing diverse processes including proteasomal degradation, DNA repair, endocytosis, and inflammatory signaling.
- As the ribosomal protein L40 component of the 60S subunit, UBA52 facilitates cap-dependent translation of a subset of cellular and viral mRNAs, such as vesicular stomatitis virus transcripts. Related references: PMID:23169626 | PMID:23636399 | PMID:32669547
- UBA52 localizes to both the cytoplasm and nucleus, supporting its dual role in ubiquitin-dependent signaling and ribosomal protein synthesis.
- The protein undergoes multiple post-translational modifications, including phosphorylation, methylation, and ADP-ribosylation, and contains zinc-binding sites critical for its structure and function.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1–100, which includes the conserved ubiquitin domain. Consequently, the antibody may detect both free ubiquitin and ubiquitin conjugates, in addition to the full-length UBA52 protein. Validate specificity in your sample system using appropriate controls.
- For Western blotting, the full-length fusion protein migrates at approximately 15 kDa. Higher molecular weight bands may represent polyubiquitinated species or cross-reactive ubiquitin–protein conjugates.
- In immunohistochemistry and immunofluorescence, expect both cytoplasmic and nuclear staining. Optimize fixation and antigen retrieval conditions for your tissue type.
- For ELISA applications, use the antibody to capture or detect UBA52 or ubiquitinated proteins, ensuring proper blocking to minimize background.
