Biological Background: UBA1/7 Function and Localization

- It activates ubiquitin via ATP-dependent adenylation of its C-terminal glycine, forming a ubiquitin-E1 thioester intermediate essential for subsequent E2/E3 transfer (PubMed:1447181). Related references: PMID:1447181

- In Western blotting (WB), expect predominant bands at ~118 kDa for UBA1 and ~112 kDa for UBA7, with possible variations depending on post-translational modifications or splice isoforms.
- For immunofluorescence (IF) and immunohistochemistry (IHC), note that UBA1 is distributed in the cytoplasm, nucleus, and mitochondria; optimize fixation and permeabilization conditions to preserve signal in these compartments.
- For ELISA applications, consider using recombinant UBA1 protein or cell lysates spiked with the target as standards, and perform cross-reactivity testing against UBA7 to confirm detection.
