Biological Background: UBA1 Function and Localization

  • UBA1 (Ubiquitin-like modifier-activating enzyme 1), also known as UBE1, is the E1 enzyme that initiates the ubiquitin-proteasome pathway by activating ubiquitin for conjugation to target proteins.
  • It catalyzes the ATP-dependent adenylation of ubiquitin's C-terminal glycine and subsequent thioester formation between ubiquitin and its active-site cysteine, releasing AMP. Related references: PMID:1447181
  • Essential for protein degradation via the ubiquitin-proteasome system, marking proteins for proteasomal degradation. Related references: PMID:1606621, PMID:33108101
  • UBA1 plays a critical role in the DNA damage response by promoting the formation of radiation-induced foci and the recruitment of repair proteins TP53BP1 and BRCA1 to damage sites, thereby ensuring timely DNA repair. Related references: PMID:22456334
  • Subcellular localization: found in the cytoplasm, nucleus, and mitochondria, consistent with its broad roles in protein homeostasis and genome stability.
  • Post-translational modifications: known to be acetylated and phosphorylated; contains nucleotide-binding and repeat motifs.
  • Ubiquitously expressed, detected in erythrocytes at the protein level; mutations in UBA1 are linked to diseases such as VEXAS syndrome and neurodegenerative disorders.

Experimental Guidance and Technical Tips

  • For Western blot, consider using a whole cell lysate and expect a band near 118 kDa.
  • For immunohistochemistry, validate staining patterns in relevant tissues, noting the known nuclear and cytoplasmic localization of UBA1.
  • For immunofluorescence, permeabilization may be required to detect nuclear signals.
  • ELISA may be used for quantification, but validate against known standards.
