Biological Background: UAP56/DDX39B Function and Localization

  • Spliceosome RNA helicase DDX39B, also known as 56 kDa U2AF65-associated protein, ATP-dependent RNA helicase p47, DEAD box protein UAP56, and HLA-B-associated transcript 1 protein, is encoded by the DDX39B (BAT1, UAP56) gene.
  • Component of the TREX complex, coupling mRNA transcription, processing, and nuclear export. Associates specifically with spliced mRNA and is recruited in a splicing- and cap-dependent manner near the 5′ end, functioning in mRNA export via the TAP/NXF1 pathway.   • Required for the first ATP-dependent step in spliceosome assembly and U2 snRNP interaction with the branchpoint. Exhibits RNA-stimulated ATP binding/hydrolysis and ATP-dependent RNA unwinding activities; the helicase and ATPase activities are not influenced by U2AF2, though ALYREF/THOC4 has a reported stimulatory effect (PMID:23299939).
