Biological Background: UACA Function and Localization
- UACA (Uveal autoantigen with coiled-coil domains and ankyrin repeats), also known as NUCLING, is a large protein with ankyrin repeats and coiled-coil domains, existing in multiple isoforms due to alternative splicing.
- It functions as a key regulator of apoptosis by controlling APAF1 expression, upregulating the apoptosome, downregulating LGALS3/galectin-3, and inactivating NF-kappa-B, thereby promoting programmed cell death under stress conditions.
- UACA also modulates isoactin dynamics to regulate cell morphology, growth, and motility; its interaction with ARF6 may influence cell shape and motility after injury, and it may participate in neurite formation.
- The protein localizes to the nucleus, cytoplasm, and cytoskeleton, reflecting its dynamic roles in both nuclear and cytoplasmic processes.
- It is highly expressed in skeletal muscle, heart, kidney, and pancreas, with additional presence in choroid, retina, epidermal melanocytes, eye muscles, and thyroid follicular cells.
- Post-translational modifications include acetylation and ubiquitination (isopeptide bond formation), and the protein features ANK repeats and coiled-coil domains essential for its interactions.
- With a molecular weight of approximately 162.5 kDa (163 kDa on SDS-PAGE), UACA is ubiquitously expressed but exhibits tissue-specific regulation.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to the N-terminal region (amino acids 1–90) of human UACA, which may contribute to recognition of both full-length and potential isoform variants; consider sequence alignment if working with highly homologous proteins.
- For Western blot, a band around 163 kDa is expected in relevant human, mouse, or rat samples; validate the antibody in your specific lysate and loading conditions, and consider using a high-molecular-weight marker.
- ELISA applications may require optimization of coating antigen and antibody dilution; the polyclonal nature can enhance sensitivity but also increase background—titration is advised.
