Biological Background: U1A/SNRPA Function and Localization
- U1A/SNRPA (Small Nuclear Ribonucleoprotein A) is a core protein component of the U1 small nuclear ribonucleoprotein (snRNP) particle, which plays a critical role in pre-mRNA splicing.
- It binds specifically to stem loop II of U1 snRNA, enabling the U1 snRNP to recognize the 5' splice site of pre-mRNA and initiate spliceosome assembly.
- In its snRNP-free form (SF-A), U1A/SNRPA may be involved in the coupling of pre-mRNA splicing and polyadenylation.
- The protein preferentially recognizes and binds to the 5'-UGCAC-3' motif on single-stranded RNAs, highlighting its sequence-specific RNA-binding activity.
- U1A/SNRPA is exclusively localized to the nucleus, consistent with its function in nuclear RNA processing events.
- Post-translational modifications of U1A/SNRPA include acetylation, methylation, and phosphorylation, which may regulate its activity or interactions.
- It belongs to the RNA-binding protein family and contains characteristic RNA recognition motifs (RRMs), as indicated by the keywords 'Repeat' and 'RNA-binding'.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1-100) of human U1A/SNRPA. This antibody is expected to detect full-length endogenous protein across the reactive species (human, mouse, rat).
- For western blotting, the predicted molecular weight is 31 kDa; a band near this size should be observed. Pre-stained markers and appropriate loading controls are recommended.
- In immunofluorescence/ICC, a nuclear staining pattern is anticipated. Co-staining with a nuclear marker or spliceosomal component can confirm specificity.
- This antibody has been used in immunoprecipitation (IP) applications; it may co-immunoprecipitate U1 snRNA and associated U1 snRNP proteins. Include RNase-free conditions if RNA interaction is to be preserved.
- As a rabbit monoclonal antibody, lot-to-lot consistency and high affinity are expected. Titration experiments should be performed for each application to determine optimal working dilutions.
