Biological Background: Tyrosine Hydroxylase Function and Localization
- Tyrosine hydroxylase (TH) is the rate-limiting enzyme in catecholamine biosynthesis, converting L-tyrosine to L-DOPA using tetrahydrobiopterin and molecular oxygen. - Alternative splicing produces multiple isoforms, some of which lack catalytic activity.
- TH is primarily expressed in the brain and adrenal glands, consistent with its role in neurotransmitter and hormone biosynthesis.
- Subcellularly, TH localizes to the cytoplasm, perinuclear region, nucleus, and is also found in cell projections such as axons and in synaptic vesicles.
- The protein undergoes phosphorylation and is associated with metal binding (iron), important for its enzymatic activity.
- Mutations in TH are linked to neurological disorders including DYT5b dystonia and Parkinson disease.

- The immunogen corresponds to the C-terminal region (aa 411–528), which may allow detection of multiple TH isoforms; validate specificity in your target tissue or cell line.
- For Western blot, a band may appear between 45–59 kDa depending on isoform and phosphorylation state; consider loading sufficient protein and using appropriate positive controls (e.g., brain or adrenal lysates).
- IHC-P and IF/ICC may require antigen retrieval; include controls and optimize detection conditions for your sample type.
- ELISA applications can be used for quantitative detection; optimize coating concentration and blocking conditions.
